DNA Barcoding of Fish Species Diversity in Guizhou, China

Guizhou is an important ecological barrier in the upper reaches of Yangtze River and Pearl basins with abundant fish species. However, from these regions are threatened by anthropogenic activities, including overfishing habitat destruction. Here, we assessed diversity more than half species region using DNA barcoding (partial sequence cytochrome c oxidase subunit I (COI) gene). We obtained 800 mitochondrial COI barcode sequences 82 genera, 18 families 8 orders fishes. The average Kimura two-parameter (K2P) distances within genera were 0.35% 5.44%, respectively. interspecific distance was 15.54 times higher mean intraspecific distance. Moreover, barcodes revealed 175 operational taxonomic units (OTUs) based on consensus demarcation schemes. Barcoding gaps detected 94.81% morphospecies. Three (Schistura fasciolata, Vanmanenia pingchowensis, Misgurnus dabryanus) have considerable variability, each divided into multiple molecular (MOTUs) definition methods (Automatic Barcode Gap Discovery, Refined Single Linkage, General Mixed Yule Coalescent, Poisson Tree Processes), possibly indicating occurrence cryptic Altogether, our study reveals complex Province, serving as a reference for conservation monitoring this region.
